Gordian is creating the first high-throughput in vivo screening platform, to extend the reach of gene therapy to complex diseases of aging.
Age-related diseases have complex etiologies that depend on interactions with the aged environment, and traditional screening methods have failed to produce effective treatments. By screening for efficacy within the most relevant animal models, our platform will identify effective therapeutics before the biology of each disease has been thoroughly mapped.
Moreover, our screening is designed to generate vast datasets with every run, even while we directly identify candidates. This integrates with our data analysis pipeline to map out the underlying biology causing disease, and even the fundamental processes of aging itself.

As our molecular biology lead, your early objectives be to design multiplexed intervention libraries, including choosing/designing promoters and reporters to limit genetic interventions to specific cell types. You will collaborate with our bioinformatics lead to design and optimize single-cell readouts for the intervention screen. By six months, you will have established the laboratory infrastructure needed to carry out these experiments, have chosen a strategy for our first intervention library, and validated expression and efficacy of this library in vitro.
Longer term, you will design multiple large-scale intervention libraries, with iterative optimization based on bioinformatic analysis of results. You will also formulate and test strategies to enrich cell populations based on their transcriptomic state. You will continue designing promoters and reporters for use in different tissues and species.
You should have the experience and hands-on technical ability to accomplish these tasks, and a mindset of continuous optimization of our platform. Experience generating, working with and analyzing multiplexed nucleotide libraries, including vector design, is crucial. Experience with multiple genetic interventions (RNAi, CRISPR and 2nd generation fusions, transgenes), with single-cell assays, and/or lab automation is a plus.
